Parental Bias in Segregation Distortion of RAPD Markers from Solanum Anther-Derived Plants

چکیده
A diploid Solanum clone 9507-04 was used to grow anther-derived plantlets that were putatively monoploid. Sixteen RAPD primers were used to observe the RAPD profiles from anther-donor and anther-derived materials. A total of 56 RAPD markers were scored and each marker was identified according to the primer’s designation and size (kilo base pairs) of the amplified fragment. Of 56 RAPD markers assayed, 44.6% did not follow the expected 1:1 Mendelian segregation (χ 2 >4.26, P<0.05) in the anther-derived progeny. The selected six morphological traits also segregated in anther-derived plants including the nine that retained 3-4 of the parental leaf characters. The 12 distortion–causing RAPD markers (χ2=8.89-17.64, P<0.003) with above 84% presence in the progeny were retained in all above nine plants. Such preference of parental markers in potato anther-derived progeny may indicate a linkage with androgenesis.

Microsatellite marker analysis of an anther-derived potato family: skewed segregation and gene-centromere mapping.
Segregation patterns of polymorphic simple sequence repeat (SSR) primer pairs were investigated in monoploid potato families derived from anther culture. A total of 14 primers developed from the sequences in the database, as well as from a genomic library of potato, was used. Distorted segregation was observed for seven (50%) polymorphic loci among monoploids derived from an interspecific hybri...

In this research, the effects of zeatin riboside (0.6, 0.8, 1, 1.2 and 1.4 mg l-1), duration of heat stress of 35 °C (6, 8, 10 and 12 days) and mannitol (0, 10, 20, 30 and 40 mg l-1) on anther culture of four eggplant cultivars were investigated in independent experiments. These experiments were performed according to a factorial arrangement in a randomized complete blocks design layout. The re...

The evolution of inbreeding is common throughout the angiosperms, although little is known about the developmental and genetic processes involved. Lycopersicon pimpinellifolium (currant tomato) is a self-compatible species with variation in outcrossing rate correlated with floral morphology. Mature flowers from inbreeding and outcrossing populations differ greatly in characters affecting mating...

Frequency of somaclonal variation in embryogenic calli and plants of Olea europaea L cv.Kroneiki derived from somatic embryogenesis were examined by randomly amplified polymorphic DNA (RAPD) analysis. Radicle and cotyledon (proximal part) of mature zygotic embryo were cultured in OMc medium supplemented with IBA (5 mgL-1) and 2-ip (0.5 mgL-1). Somatic embryogenesis was induced in calli of radic...
متن کاملGlycoalkaloid profile in potato haploids derived from solanum tuberosum-S. bulbocastanum somatic hybrids.
Cultivated and wild potato species synthesize a wide variety of steroidal glycoalkaloids (GA) that may affect either human health or biotic stress resistance. Therefore, GA composition must be a major criterion in the evaluation of breeding products when species genomes are merged and/or manipulated. This work reports the results of GA analysis performed on unique haploid (2n=2x=24) plants obta...
